NEW MATERIAL:A human calpastatin polypeptide expressed by the formula. EXAMPLE:A remedy for diseases caused by an overaction of a calpain, such as adult leukemia, cataract, etc. PREPARATION:For example, a cDNA prepared from a template mRNA originating from a normal human liver is linked to a vector and introduced into a host cell to form a cDNA library. A clone containing a gene coding a human calpastatin is screened from the cDNA library by using a cDNA fragment of a porcine calpastatin as a probe. The clone is then cultured and propagated and a bacteria pellet collected is treated with restriction enzymes to take up the DNA fragment, which is inserted into a vector and introduced into E. coli to transform. The resultant transformant is then cultured and a peptide expressed by the formula is obtained from the culture solution. |
