A purified, highly-active profibrinolysin substantially free of inhibitors is obtained by suspending the profibrinolysin in aqueous acid of pH 1 to 5, separating the acid solution from the insoluble contaminating material, neutralizing the acid solution until the pH is between 6 and 8 and adding a water-soluble salt of cadmium, mercury, lead, or zinc to precipitate the profibrinolysin from the solution in the form of a metal salt. Profibrinolysin is decomposed by (a) dialysis against distilled water to remove the metal, the dialysate being lyophilized, (b) treatment with an ion-exchange resin having free acid groups, e.g. Dowed 50 (H form), followed by lysphilization of the supernatant, or (c) treatment with disodium ethylenediamine tetraacetic acid followed by dialysis and lysphilization of the dialysate. The profibrinolysin may be obtained from human or mammalian blood such as borine blood. Specification 840,192 is referred to.
