<p>PURPOSE:To cultivate an adhesive animal cell in a floated state stably for a long period without using a carrier by limiting Ca<2+> concentration in a growth phase and maintenance phase and multiplying and maintaining the adhesive animal cell in a serum-free medium. CONSTITUTION:In multiplying and maintaining an adhesive animal cell in a serum-free medium, concentration of calcium ion in a culture system is made into <0.3mM, preferably 0.001-0.3mM, further preferably 0.02-0.3mM at one period in growth phase and maintenance phase. The average number of cells becomes 1-15 based on 1 lump of cell coagulation and cells in the interior of cell lump can be multiplied and maintained stably for a long period of time without being necrosed by limiting the concentration of calcium ion the culture system. Strain 293 derived from human fetal nephrocyte, BHK cell, CHO cell, COS cell, rat HepI cell, human lung cell, mouse hepatocyte, etc., are preferable as the adhesive animal cell.</p> |
