NEW MATERIAL:A heat-resistant lipoprotein lipase having the following properties. Activity, acting to triglyceride in lipoprotein and hydrolyzing to glycerol and fatty acid; heat-resistance, completely maintaining the original activity after the treatment in a buffer solution at 60 deg.C for 15min; ratio of glycerol- forming activity, >=15% based on fatty acid-forming activity. USE:A quantitative determination reagent for triglyceride in blood. Only a small amount of the reagent is required because of its high thermal stability and high glycerol-forming activity. PREPARATION:The objective enzyme can be separated and purified from a culture product of a thermophilic actinomycete [e.g., Streptomyces 7825 (FERM P-9983)] e.g., by removing the microbial cells from the culture liquid and subjecting the filtrate to hydrophobic chromatography. |
