NEW MATERIAL:A factor I consisting of a protein produced by R-1 clone cells of myelogenous leukemia M1 cells, capable of inhibiting differentiation of myelogenous leukemia cells and showing simple band at about 68000 molecular weight in the SDS-polyacrylamide gel electrophoresis. USE:A remedy for myelogenous leukemia capable of inhibiting differentiation of myelogenous leukemia cells. PREPARATION:For example myelogenous leukemia M1 cells established from spontaneously myelogenous leukemia of a SL mouse are cultured and R-1 clone cells of resistant M1 cells free from differentiation-induction into mature cells are isolated from the parent strain sensitive to differentiation-inducing substances. The resultant R-1 clone cells are cultured on a culture medium and a factor I is separated from the culture solution by the ammonium sulfate preparation and purified by the chromatography using CM-Sepharose CL-6B column and Sephadex G-200 column in order. The purified factor I is further purified by the reverse phase high-performance liquid chromatography using 18-C hydrophobic carrier and by the high-performance liquid chromatography using a gel filtration column, thus obtaining the factor I. |
