The present invention relates to a method and to a device for the qualitative and/or quantitative determination of a ligand present in a fluid, the determination being very simple to carry out and, where appropriate, easy to automate. The device according to the invention comprises, in combination: - means for bringing the test fluid sample F and the reactants A, B and C into contact; - a first reaction zone Z1, arranged to receive a test fluid sample F and at least one liquid or solid labelled reactant A capable of recognizing at least one ligand L of interest possibly contained in the abovementioned sample, to form a soluble L-A complex; - a second reaction zone Z2, separate from the first, comprising a solid phase 21 containing an antigen and/or an antibody and/or a complement of DNA and/or a complement of RNA capable of reacting only with the labelled reactant(s) A not bound to the ligand L, to form, where appropriate an insoluble A-B complex; - a third reaction zone Z3, which contains a third reactant C for visualising the presence of the L-A complex, and, where appropriate - means 32 for reading the reaction visualised in the zone Z3; - means for automation of the sequential operation of the aforementioned means for bringing into contact and reading means. n
