PURPOSE: To obtain red blood cells which are converted to an H-antigen type to be used in the transfusion therapy by equilibrating B or A, B antigen type red blood cells to a specific pH, bringing the red blood cells into contact with α-galactositase to remove galactositase and equilibrating its pH to a specific value. CONSTITUTION: B or A, B antigen type red blood cells are equilibrated to a pH between 5.7-5.8 and the cells are brought into contact with α-galactosidase for a time enough to convert the B antigen to the H antigen in the red blood cells and the α-galactosidase is removed. Then, the treated red blood cells are equilibrated again to a pH of 7.2-7.4 whereby the objective directly transfusable O- or AH-type red blood cells which i) have no P1 antigenicity, ii) absorb and release oxygen in vivo, iii) have higher content of adenosine-5'- triphosphate than 90% content of naturally occurring B type red blood cells, iv) substantially contain no terminal chain-α-linked galactose trihexosylceramide and v) contain no type B antigenicity according to the analysis with the red blood cell coagulation reaction, are obtained. |
