<p>PURPOSE: To provide a method for storing an embryo at a low temperature, enabling to effectively store at the low temperature the embryo useful for freezing the mammalian embryo for establishing the low temperature pack of the mammalian embryo in low temperature biology, by cooling a buffer salt solution containing a specific compound as an antifreezing agent to a cooling medium temperature. CONSTITUTION: A buffer solution containing an antifreezing agent mixture comprising one of glycerol, dimethyl sulfoxide and a dihydric alcohol derivative such as 1,2- propanediol and further a biologically active substance such as a 0.4% concentration choline chloride solution is cooled in a highly heat-diffusing powdery substance, such as zinc, preliminarily cooled to the temperature of a cooling medium such as liquid nitrogen to store the embryo at the low temperature. This method is useful for the low temperature freezing of a low temperature bank used for the embryo of a genetically valuable rat race, etc., especially used for advance of experiment-clinical medicine as an antisterility means and for the advances related to the progress of livestock industry for reinforcing the genegiology of a highly valuable animal. The method enables to decrease the number of operations and reduce a time for carrying out all the cycles of the low temperature storage.</p> |
