C-terminal amidated peptides are prepared by preparing the corresponding C-terminal Gly-extended peptide by solid-phase synthesis and converting enzymatically, typically using peptidylglycine alpha -amidating monooxygenase (PAM), the resulting Gly-extended peptide to the corresponding des-glycine peptide amide.
