<p>Dideoxy DNA sequencing method using the substantially random insertion of the gamma-delta element of an F-plasmid into a starting plasmid containing the DNA to be sequenced. Fragments comprising a portion of the DNA to be sequenced and, adjacent thereto, either the gamma end or the delta end of the gamma-delta insert are isolated and subjected to dideoxy sequencing using an oligonucleotide having a length of at least 10 nucleotides, which is complementary to both 5' ends of the gamma-delta element, as a primer.
