<p>PURPOSE:To obtain a large amount of the subject compound in high efficiency by carrying out the tissue-culture of a tissue or cell of a specific plant in a medium composed of an oligopeptide containing phenylalanine. CONSTITUTION:Tissue of a plant capable of producing tropane-type alkaloid such as a plant of genus Duboisia (e.g. Duboisia myoporodies) is tissue-cultured to obtain an adventitious root (A). Separately, a tissue culture medium (B) is prepared by compounding 0.5-500muM of an oligopeptide of formula (R is amino acid residue; n is 1-5) (e.g. triphenylalanine) containing phenylalanine as a constituent amino acid, Murashige-Skoog medium, 1-100g.l of a carbon source (e.g. sucrose) and 0.01-100muM of plant hormones, etc. (e.g. indoleacetic acid). The component A is inoculated in the medium B and cultured for about 3 weeks. The obtained adventitious root is dried, extracted with chloroform and purified to recover the objective tropane-type alkaloid.</p> |
