<p>PURPOSE:To use a monoclonal antibody as a diagnosticum of infectious diseases by immunizing a cell against an attenuated strain of bovine infectious pathogenic virus to give sensitized B lymphocyte, selecting the B lymphocyte as a hybridoma and cultivating. CONSTITUTION:An attenuated strain of bovine infectious pathogenic virus is multiplied in cell HmLu-1 (A) derived from hamster fetus cultivated in a serum- free medium to give a culture solution of virus (B). Then Akabane virus (ADA), bovine infectious pathogenic virus is adsorbed on the cell A and cultivated to give a culture mixture, which is purified to give a purified product of virus part (C). Then hybridoma culture is carried out by using the culture solution B as an immune source for Akabane virus and the component C as an immune source for Ibaraki virus to give hybridoma cell (D), respectively. Then the cell D is cloned, cultivated in a medium containing growth factor, etc., to give a culture mixture, which is purified by salting-out, etc., to produce a monoclonal antibody such as 4F-12 or 8B-9 to be specifically reacted with a bovine infectious pathogenic virus.</p> |
