DOWN'S DISEASE DIAGNOSING REAGENT FOR SERUM

PURPOSE:To diagnose a Down's disease by using a monoclonal antibody which conjugates with the Chinese hamster ovary cells (CHO cells) synthesizing the membrane protein coded from the human chromosome No.21 and does not conjugate with the normal CHO cells. CONSTITUTION:Sample serum is subjected to gel electrophoresis then to electroblotting to transfer the protein in the gel to a nitrocellulose film. This film is treated with a blocking liquid and is then immersed in the soln. of the monoclonal antibody which conjugates with the CHO cells synthesizing the membrane protein coded from the human chromosome No.21 and does not conjugate with the normal CHO cells. Further, the cells are brought into reaction in a biotinated anti-mouse IgG soln. and an avidin biotinated western horse radish peroxidase complex after cleaning is added thereto to effect reaction. This nitrocellulose film is immersed in a color forming liquid so that the protein band forms colors. The protein of 40 kilodalton existing specifically in the serum of the Down's disease patient is specifically detected in this way.
