PURPOSE:To detect the enzyme in a specific enzyme reaction system with high sensitivity by using an amperometric detector. CONSTITUTION:A liquid feed pump 2, an auto injector 3, a column 4, and a detector 5 are connected in this order from a moving phase supply part to a detecting system 1. Further, a high performance liquid chromatograph device constituted of a sample analysis flow passage (a) extended to a drain, a control section 6 added to the flow passage (a) and a recorder 7 is used. The prescribed enzyme reaction is progressed by using a water soluble magnesium salt of oxygen acid anion having a high oxidation number in the central element for the enzyme reaction system essentially consisting of the magnesium ions to be detected in the detection method. The reaction mixture is thereafter subjected to the electrochemical oxidation by electrolysis and the resulted product of the enzyme reaction is detected by a prescribed electrolytic current value. The enzyme of the enzyme reaction system is detected with the high sensitivity in accordance with the result of the detection. |
