PURPOSE:To enable diagnosis of cancerous diseases, etc., by subjecting a specific cell and a myelomatous cell to cell fusion and culturing the resultant hybridoma. CONSTITUTION:An N-acetylneuraminic acid derivative having a beta-configuration expressed by the structural formula is used to immunize a Balb/c mouse and afford (a) a B cell (lymphocyte), which is then fused to a myelomatous cell that is a myeloma cell derived from a human, etc., in the presence of polyethylene glycol to provide (b) a hybridoma. The resultant cell (b) is then cultured in an HAT culture medium, etc., to afford a culture supernatant, which is subsequently screened by an enzyme antibody method, etc., to produce a monoclonal antibody having specificity for N-acetylneuraminic acid having the beta-configuration as an epitope (antigenic determinant). The antibody, as necessary, is used to diagnose blood serum or urine of malignant tumor or acute inflammatory diseases. |
