<p>PURPOSE:To make possible to develop of vaccine and diagnostic, etc., of AIDS virus by determining whole base sequence of DNA exhibiting complementarity to gene RNA of immunodeficient virus of monkey. CONSTITUTION:DNA exhibiting complementarity of gene RNA of immunodeficient virus of monkey (SIVAGM) is subjected to isolation and cloning. In the whole sequence of DNA exhibiting complementarity to RNA of SIVAGM, an existence of EcoRI site is known as a restricted enzyme site at only one position, so said closing is carried out utilizing said restricted enzyme site. Next, lambda phage DNA in which said DNA is integrated is constructed and said lambdaphage DNA is subjected to sub-cloning using transformable Escherichia coli. For instance, lambdagtwES SIVAGM is digested by restricted enzyme EcoRI and resultant fraction is integrated to plasmid containing same restricted enzyme site. Finally, Escherichia coli is transformed by resultant plasmid.</p> |
