DNA CONTAINING BIOSYNTHETIC GENE GROUP OF BETA-LACTAM ANTIBIOTIC

PURPOSE:To improve production of a specific DNA, by transducing a bonded substance of chromosome DNA and a plasmid vector to a variant deficient in isopenicillin N synthase (IPNS) activity. CONSTITUTION:Chromosome DNA is extracted from a cell of Streptomyces lipmanii and treated with a restriction enzyme to give DNA fragment (A) of 9-20Kb. Then Streptomyces lipmanii is varied by ultaviolet light irradiation, etc., a strain not to produce isopenicillin N is separated and selectively recultivated to give 1PNS deficient activity strain (C9504) (C). A bonded substance of the fragment A and the component B is transduced to the strain C to give a transformant (D). The cell D is extracted and purified to produce a DNA containing a biosynthetic gene groups of beta-lactam antibiotic derived from Streptomyces lipmanii capable of being complementary to variation of 1PNS activity deficient variant and/or a variant not to produce beta-lactam antibiotic.
