<p>Nucleoside phosphoramidites have been synthesized and subsequently utilized in the preparation of tritium-labeled nucleotides at predetermined sites of polycnucleotides or oligonucleotides. Purine and pyrimidine nucleotides labeled in specific positions with tritium are protected at the reactive sites and then condensed in a predetermined sequence to yield tritium-labeled polynucleotides. The tritium-labeled oligonucleotides may be used to determine the effects of altering the primary DNA structure on the formation and repair of DNA adducts generated by chemical carcinogens and anticancer drugs.</p> |
