摘要 |
<p>PURPOSE:To select a cell producing a required protein and collect the cell as a high-purity clone, by labeling a protein relating to selectable phenotype with fluorescence and quantitatively determining the labeled protein. CONSTITUTION:A animal cell is subjected to contrasformation with an external DNA I and an external DNA II coding a selectable phenotype which is not manifested essentially with said animal cell. The transformed cell is cultured in the presence of a selection reagent to amplify the external DNA II coding the selectable phenotype. The cotransformant is made to contact with a fluorescent labeling substance reactive with the gene translation protein of the external ENA I and/or external DNA II and is quantitatively determined by using fluorescence as a criterion. A clone producing a gene translation protein of external DNA I can be efficintly selected and collected from the cotransformant by this process.</p> |
