PURPOSE:To analyze an oil substance in a short time in a high sensitivity, by amperometrically measure the oily substance with an oxidase-immobilized enzyme electrode in an organic solvent having the solubility of water in the solvent in a specific range and electrically conductible when a supporting salt is dissolved in the solvent. CONSTITUTION:A working electrode 1 whose surface is covered with an enzyme electrode 2 having an immobilized oxidase (e.g., cholesterol oxidase) thereon is immersed in one side of an H type cell 5 filled with an organic solvent 6 having a water solubility of <10wt.% (at 20 deg.C) and electrically conductible when a supporting salt is dissolved in the solvent. In the other side of the H type cell 5, an organic solvent 7 mixed with a supporting salt is filled and a reference electrode 3 and a counter electrode 4 are immersed. These are connected with a potentiostat 10 and a recorder 11 and held at 40 deg.C in a constant temperature bath. An oily substance to be measured is added to the one side of the H type cell 5 to allow the oily substance to enzymatically react with oxygen. The produced H2O2 is amperometrically measured with a potentiostat 10 from the change of electric currents under a constant voltage to determine the concentration of the oily substance. |
