REVERSE PHASE CHROMATOGRAPHY IN OLIGONNUCLEOTIDE SEQUENCING

A method for determining oligonucletide sequence is presented. Thus, as shown in Figure 1, 4 Erlenmayer flasks are connected into C18Sep- pak reverse phase chromatography. G-cartridge is soaked into dimethyl sulfate solution for 2 min. A-cartridge is treated with 50% formic acid for 4 min. T-cartridge and Ccartridge are immersed into 60% hydrazine solution respectively for 3 min and 4 min. The properly cut DNA segments are eluted from the column and the sample is analysed automatically to get nucleotide sequence.
