Labelled antibodies against reagin Ig or against fragments of reagin Ig are used to determine reagin Ig (reagins are antibodies against allergens). The antibodies may be labelled with a radioactive isotope, for example I<SP>125</SP>, or with a fluorescent group using for example fluorescein isothiocyanate. Means for determining reagins comprises a first reagent comprising a water-insoluble polymer having bound to it allergen against which the reagin to be determined is directed and a second reagent comprising the labelled antibodies as above. The sample is incubated with the first reagent when any reagin present in the sample becomes attached to the polymer particles; the polymer particles are separated from the liquid phase, washed and then incubated with the second reagent. The solid and liquid phases are separated again and the radiation in either phase determined and compared with that in a control. The water-insoluble polymer is preferably particulate but may be in the form of discs, strips or a test tube wall and may be a copolymer of a polyhydroxy compound for example dextran, starch, dextrins, polyvinylalcohol with a bifunctional substance. The allergen may be physically or chemically bound to the polymer. Examples relate to Sephadex (RTM) as carrier, birch pollen as allergen and I<SP>125</SP>-labelled rabbit antibody.</p> |
