<p>PURPOSE:To industrially and advantageously obtain the title compound for fibrinolytic agent, etc., by solubilizing a product of enzyme inserting a plasmid having variant DNA sequence in epidermal growth factor domain of human prourokinase using guanidine hydrochloride. CONSTITUTION:An enzyme transformed by a plasmid integrating DNA sequence coding a valiant human prourokinase losing total domain or part of epidermal growth factor domain of human prourokinase or replacing the total domain or part with other amino acid residue is cultured in a culture medium and then yeast cell is collected, homogenized and centrifuged to recover a precipitated fraction, to which 6M guanidine hydrochloride (pH9.0) is added to solubilize the precipitated fraction and the resultant supernatant is treated with a buffer for reconstruction and then purified by an affinity chromatography to provide the aimed compound.</p> |
