Orotidine - 5<SP>1 </SP>- monophosphate and/or uridine - 5<SP>1 </SP>- monophosphate, specifically or unspecifically labelled with a radioisotope, are prepared by incubating the corresponding nucleic acid base (together with 6-azauridine- 5<SP>1</SP>-monophosphate when the base is orotic-6- <SP>14</SP>C acid) in the presence of 5-phosphoribosyl-1- pyrophosphate or its metabolic precursors, in the medium of a buffer and Mg<SP>2+</SP> ions at 27‹ to 37‹ C. in the presence of the unfractionated supernatant fraction of the cell culture sonicate of Brevibacterium Ammoniagenes (strain 364 of the Culture Collection of Entomogenous Bacteria, Prague), and isolating the reaction product by paper chromatography, or after adjusting the enzymatic system, the product may be used as the source for the synthesis of the corresponding triphosphates. The radioisotope is preferably <SP>14</SP>C or <SP>3</SP>H. |
