<p>PURPOSE:To easily separate the insoluble fractions of crushed cells by repeatedly filtering the crushed cells plural times with an ultrafilter membrane without cleaning the membrane. CONSTITUTION:The liquid suspension of the crushed cells obtd. by culturing the colon bacilli holding the plasmid incorporated with, for example, human prourokinase gene and suspending the bacteria into a tris buffer soln., then crushing the soln. with a 'Gaulin(R)' homogenizer is concentrated by using the ultrafilter membrane to remove the soluble materials in the bacteria. The operation to further add the buffer soln. to the suspension then to concentrate the same with the ultrafilter membrane is then repeated several times and the residues of the crushed cells are cleaned. Since the permeation flow rate is restored by the dilution with the buffer soln., the membrane cleaning stage is not needed between a series of concentrating stages. The separated soluble materials or insoluble fractions are recovered and utilized at need.</p> |
