NOVEL CELLULAR STRAIN MS128 CAPABLE OF PRODUCING HEPATITIS B VIRAL SURFACE ANTIGEN AND PRODUCTION OF HEPATITIS B VIRAL SURFACE ANTIGEN CONTAINING PRE S2 PROTEIN

摘要

PURPOSE:To obtain a strain MS128 capable of producing hepatitis B viral surface antigen, by inserting a DNA fragment free from a C gene part of a specific hepatitis B viral DNA into a plasmid and transfecting the resultant expression vector into a prescribed cell. CONSTITUTION:A DNA fragment free from a C gene part of a subtype adr type hepatitis B viral DNA is inserted into a plasmid to synthesize an expression vector, which is then transfected into a cellular strain C127 (ATCC CRL1616) derived from an R-III mouse to transform the cellular strain. The obtained transformant is subsequently cloned to afford a cellular strain having a spindle- like form. The resultant cellular strain is then cultured by a conventional method to extract and purity the aimed pre S2 protein and hepatitis B viral surface antigen from the culture supernatant liquid.
