PURPOSE:To obtain a novel urease with a specific activity of 20U/mg protein at 37 deg.C and optimum pH, by culture on a culture medium urease-producing bacteria classified as Lactobacillus or Streptococcus. CONSTITUTION:Acidic urease-producing bacteria classified as Lactobacillus or Streptococcus, e.g., Lactobacillus fermentum. Lactobacillus reutori, Streptococ cus bovis is put to culture in a culture medium to accumulate urease in a culture product followed by collection of the urease. This urease acts best on urea, being 1.5-5.6 in the optimum pH and stable in the pH range of 6-8 when treated at 37 deg.C for 20min. Its molecular weight determined by the gel permeation tech nique is 100,000-250,000, and its specific activity at 37 deg.C and the optimum pH is >=20U/mg protein. |
