1308022 Benzthiazine derivatives RECORDATI SA CHEMICAL & PHARMACEUTICAL CO 8 May 1970 [9 May 1969] 22395/70 Heading C2C Novel compounds I in which one of X and Y is CH 2 and the other a single bond and R is H 1 alkyl, alkenyl, propargyl, benzyl, ethoxycarbonyl, dialkylaminoalkyl, cycloaminoalkyl, CH 2 COOR 1 (R 1 is H, alkyl, dialkylaminoalkyl or cycloaminoalkyl), CH 2 CONR 2 R 3 (R 2 and R 3 are H, alkyl, alkenyl, dialkylaminoalkyl or cycloaminoalkyl or NR 2 R 3 is a heterocyclic group) or phenyl optionally substituted by chloro or sulphamyl are prepared by (1) condensing RNH 2 with a compound II (2) cyclizing a compound III or IV with the loss of H 2 O or RNH 2 respectively, (3) hydrolysis of a compound V (4) substituting a compound I where R is H with a compound capable of replacing H by a different R value, (5) reacting a compound I where R is CH 2 COOH with HN(R 2 )R 3 to give a compound I where R is CH 2 CONR 2 R 3 or (6) reacting a compound I where R is CH 2 COOH with R 1 OH to give I where R is CH 2 COOR 1 . Intermediates isolated are (A) o-cyanomethylbenzene-sulphonyl chloride, prepared by diazotization of o-aminophenyl-acetonitrile followed by reaction with SO 2 ; (B) o-cyanomethyl-benzenesulphonamide, prepared by reaction of (A) with NH 3 ; compounds VI prepared by reaction of (A) with the appropriate amine RNH 2 ; compounds VII prepared by hydrolysis of compounds VI; (C) potassium o-cyanomethyl-benzene-sulphonate, prepared by reaction of the sulphonyl chloride with KOH; (D) potassium o-carboxymethyl-benzene sulphonate, prepared by hydrolysis of (C); compounds V, prepared by oyclization of compounds VI; compounds VIII prepared by reaction of o-cyano-benzyl-sulphonyl chloride with RNH 2 ; compounds IX prepared by hydrolysis of compounds VIII; (E) sodium 2-carboxy-benzylsulphonate, prepared by hydrolysis of the appropriate 2-cyano derivative; (F) o - chlorocarbonyl - benzyl sulphochloride, prepared by reaction of (E) with PCl 5 ; (G) o - carbamyl - benzyl sulphonamide, prepared by reaction of (F) with NH 3 and (H) 3,4-dihydro- 4-imino-1H-2,3-benzthiazine-S-dioxide, prepared by cyclization of o-cyano-benzyl-sulphonamide. |
