PRODUCTION OF ERYTHROPOIETIN

PURPOSE:To enable isolation of erythropoietin of high purity by conducting adsorption and elution using monoclonal(MoC) antierythropoietin(EPO) antibody which is obtained by giving a specific hybridoma. CONSTITUTION:Hybridoma cells (HB) are obtained by effecting cell fusion between spleen cells of mouse or the like, immunized with antigen EPO of higher than 60wt.% purity and myeloma cells. The HB cells are screened by bonding it to <125>1-EPO to obtain anti-EPO antibody-producing HB. The antibody- producing HB is given peritoneally to a mouse to produce the antibody, then the ascites is fractionated with ammonium sulfate and columns to prepare monoclonal anti-EPO antibody. The sample containing EPO is adsorbed on an adsorbent to which the EPO antibody is bonded, such as affinity gel 10, and the EPO is eluted a mixed solution of acetic acid and sodium chloride to collect erythropoietin of high purity.
