PURPOSE:To obtain the title enzyme without forming by-products by cultivating a fungus belonging to the genus Rhodotorula, capable of producing hydroxamic acid hydrolase. CONSTITUTION:A fungus (e.g., Rhodotorula graminis KSM-18). capable of producing hydroxamic acid hydrolase is inoculated into a medium containing 0.5-5.0wt.% carbon source (e.g., glucose), 0.054-0.4wt.% nitrogen source (e.g., hydroxylamine) and 0.1-0.5wt.% acethydroxamic acid at pH5.0-8.0, aerobically cultivated at 25-30 deg.C for 2-10 days, the cell is ground and centrifuged to give hydroxamic acid hydrolase having amidase activity. Then a hydroxamic acid shown by the formula (R is aliphatic or aromatic group which may contain substituent group; RCO is amino acid residue) is hydrolyzed with the hydrolase in a buffer solution at pH6-10 at 0-30 deg.C to give a carboxylic acid. |
