PURPOSE:To readily change a gene coding cyclomaltodextrin glucanotransferase (CGTase) to a gene coding a gene having enhanced amylase activity by losing a required part of carboxy end side of the gene coding CGTase. CONSTITUTION:A gene coding cyclomaltodextrin glucanotransferase (CGTase) which is a cyclodextrin-producing enzyme is prepared. The gene is treated by a method reacting the gene with a restriction enzyme, method reacting the gene with exonuclease III and mung bean nuclease used together, etc., and a required part of the carboxy end side of the gene is lost by the above- mentioned treatment to change the gene to a gene coding enzyme having enhanced amylase activity. As the lost part, a part in the range from carboxy end side of the gene coding CGTase to <=600 pair of bases is preferable. |
