PURPOSE:To advantageously provide the subject component itself soluble in water and useful as a reagent capable of readily 9- fluorenylmethoxycarbonylating an amino acid, etc., in an aqueous solution, by reacting a phenolic compound with a halide compound in the presence of a specific amine. CONSTITUTION:A phenolic compound of formula I is reacted with a halide compound of formula II (R is 9-fluorenylmethoxy group; X is halogen) in the presence of a tertiary amine, particularly preferably triethylamine to provide a compound of formula III. The compound of formula III itself is soluble is water and is readily capable of 9-fluorenylmethoxycarbonylating specimens originated in living body such as amino acids, peptides or proteins in aqueous solutions under mild conditions. Since the acylation is a reaction in an aqueous solution, it is possible that only the alpha-amino group or side chain amino group of the amino acid or peptide is selectively reacted with the acylating agent by adjusting the pH of the aqueous solution. |
