PURPOSE:To efficiently produce a high-quality peroxidase, by culturing tissual pieces or cells from plant bodies of Zoysia japonica and/or Zoysia macrostachya in a nutrient culture medium and isolating the peroxidase from the resultant culture. CONSTITUTION:Cell groups or tissual pieces, such as root, hypocotyl or cotyledon of seedlings or root, stem or leaf of mature plants, from plant bodies of Zoysia japonica and/or Zoysia macrostachya are collected and aseptically cultured using a suitable culture medium to induce calli. The induced calli are then subcultured in a solid or liquid culture medium and separated into the calli and culture medium components by filtration or centrifugation, etc. The calli are subsequently homogenized in a suitable solvent to remove solid substances and provide an extract, which is then salted out with ammonium sulfate. Inorganic ions are subsequently removed by dialysis to produce a crude enzymic preparation of a peroxidase, which is subsequently purified. |
