<p>PURPOSE:To readily cultivate a large amount of Piricularia oryzae and to produce a large amount of an antibiotic, by inoculating Piricularia oryzae into a liquid culture cell of rice plant and cultivating. CONSTITUTION:Seeds of rice plant are sterilized with ethanol, etc., laid on N6 agar medium containing a carbohydrate, CaCl2, nicotinic acid, etc., a callus obtained by callus induction and subculture is subjected to shaking culture in the N6 liquid medium at 20-30 deg.C for 2-3 weeks to give a liquid culture cell (A) of rice plant. Then, Piricularia oryzae is subjected to subculture in PAD medium (e.g., potato agar medium), transplanted to PAD medium containing decocted solution of rice straw, cultivated at 26-28 deg.C for 10-14 days to give a moss layer, which is filtered to give a solution (B) containing spore of Piricularia oryzae. Then, the solution B is inoculated to the cell A and tissue culture is carried out at about one day to give a culture mixture, which is extracted with methanol, etc., and purified to recover an antibiotic such as momilactone or oryzaloxin.</p> |
