FORMATION OF POLYHEDRON BY MULTIPLYING NUCLEAR POLYHEDRON VIRUS OF INSECT BY USING INSECT CELL

PURPOSE:To readily and inexpensively improve industrial mass production of a polyhedron useful as an expellent for insect pests in the field of agricultural chemical, by infecting an insect cell cultivated in a specific medium with a nuclear polyhedron virus of insect. CONSTITUTION:1-8g/l yeast powder such as baker's yeast is blended with 3-15g/l albumin hydrolyzate, NaCl, KCl, MgSO4, CaCl2, saccharide, etc., and adjusted to pH5.6-7 to give a medium of essential components. Then, the medium is sterilized in an autoclave under 1-1.5 atmospheric pressure for 15-30minutes at 15-130 deg.C to give a medium in a sterile state. Then, the medium is dividedly poured to each plastic flask (25cm<2> bottom area) in an amount of 4-5ml, cell SES MaBr.4 derived from fat of cabbage armyworm is inoculated into the medium, a virus suspension derived from a nuclear polyhedron having about 32 unit titers is added so as to give about 4 units and the mixed solution of the virus and the cell is subjected to stationary culture at about 25 deg.C for about 7 days to multiply nuclear polyhedron viruses of insect in the insect cell and to form a polyhedron.
