<p>PURPOSE:To obtain the title line good in yield, growth rate, uniformity and disease resistance without the need for emasculation operation, by conjugating the cytoplasm of cytoplasm male sterility line and the nuclei from the protoplast of normal line origin under the protoplast-fusing conditions. CONSTITUTION:Carrot seeds of cytoplasm male sterility line are sterilized and sown on a culture medium to effect germination. Each 5mm of the lower part of the hypocotyl of the resulting sprouts is cut out and transplanted to the culture medium to form cell followed by subculture and then the protoplast is isolated in the dark followed by irradiating the protoplast with X-rays to deactivate the nuclei. In a similar way, the protoplast is prepared from the seeds of carrot with pollen fertility and is mixed with the former protoplast followed by conjugation between the two kinds of protoplast under their fusing conditions using e.g., polyethylene glycol, thus obtaining the objective novel cytoplasm male sterility line.</p> |
