<p>PURPOSE:To provide the title efficient extraction process with enhanced recovery and specific activity, by extract bacteria in the presence of a higher fatty acid sorbitan ester-polyethylene oxide condensate. CONSTITUTION:Microorganisms (e.g., yeast) transformed with a recombinant vector into which hepatitis B virus surface antigen (HBsAg) gene has been incorporated in the gene manipulation is subjected to culture in a culture medium to obtain bacterial (a) in which HBsAg has been manifested and produced. Thence, a buffer solution such as a tris buffer solution with a pH of 7-9 is spiked with 0.1-2.0W/V% of a higher fatty acid sorbitan ester-polyethylene oxide condensate (b) (e.g., monolauric acid sorbitan ester-polyethylene oxide condensate) followed by adding and grinding the bacteria (a) to extract HBsAg therefrom, thus obtaining a HBsAg extract fraction (C). The fraction C is then further fractionated and purified by e.g., an absorption chromatography using a colloidal silicate, thus isolating the objective hepatitis B virus surface antigen.</p> |
