<p>PURPOSE:To make it possible to continuously carry out culture having high concentration of an animal cell, by adding a coagulant consisting of sodium polyacrylate and chitosan into a culture tank and simultaneously feeding a fresh culture medium and discharging an old culture medium. CONSTITUTION:0.005-0.01wt.% sodium polyacrylate being an anion based high polymer coagulant and having 2.2-7.0X10<4> polymerization degree and 0.014-0.02wt.% chitosan are added into a culture medium to coagulate a cell. Then the coagulated cell is separated from the culture medium and the cell is accumulated in a culture tank and old culture medium is discharged out of the tank and a fresh culture medium is fed into the tank. When hybridoma is cultured using a serum-containing culture medium, the hybridoma can be readily cultured also in increased antibody production amount. According to the above-mentioned method, perfusion culture can be continuously carried out by a cell having high concentration. Further, scale-up is readily carried out, because the above-mentioned method is readily operatable compared with a conventional method and also complicate device is not required.</p> |
