<p>PURPOSE:To enable the mass production of the titled factor useful for the remedy of thrombosis, etc., in high safety, by linking a specific DNA sequence with a sequence of other gene, transforming a cell with a DNA having the above DNA sequence, and culturing the transformed cell. CONSTITUTION:A TPA expression vector is produced by linking (A) the sequence of the promotor region of the thymidine-kinase gene of simple herpes virus or initial gene of SV40 of other functioning gene to (B) the 5'-terminal of the DNA sequence containing the sequence from the codon ATG of the translation-initiation amino acid methionine to the terminal codon TGA of the chromosome coding the human tissue plasminogen activation factor of figure. The vector is introduced into the established cell originated from blood cell (e.g. lymphoblastocyte) to obtain a transformed cell, which is cultured in a serum-free medium, etc., to obtain TPA.</p> |
