摘要 |
PURPOSE:To efficiently produce an alkaline protease by introducing a DNA fragment containing a structural gene capable of coding an alkaline protease derived from a microorganism of the genus Bacillus into a microorganism and cultivating the resultant transformed microorganism. CONSTITUTION:A DNA fragment which is a DNA fragment containing a structural gene capable of coding an alkaline protease derived from a microorganism of the genus Bacillus and a region having both ends defined by cleavage points by restriction enzymes HpaI and HaeIII or a shortened DNA fragment within a range so as not break expression of the above-mentioned structural gene is prepared. The resultant DNA fragment is then introduced into a microorganism to form a transformant, which is subsequently cultivated in a culture medium to collect an aline protease from the culture. Purification is then carried out to afford the aimed alkaline protease. |
