PURPOSE: To improve the sensitivity for measuring theophylline by setting the concentration of anti-theophylline to a value for reducing the enzyme activation of enzyme/theophylline binding body under the non-presence of theophylline. CONSTITUTION: A sample that is predicted to contain theophylline, enzyme/ theophylline binding body, and anti-theophylline generated from theophylline that is bound to an antigen at 3-order are mixed in a water-based medium. At this time, anti-theophylline concentration is set to a concentration for reducing the enzyme activity of enzyme/theophylline binding body essentially under the non-presence of theophylline. Then, the operation of the sample for the enzyme activity of the enzyme/theophylline binding body is analyzed.
