PURPOSE:To produce a novel pullulanase having high optimum temperature of enzymatic reaction and stable pH range in weakly acidic or weakly alkaline region, by culturing a thermophilic microorganism belonging to Thermus genus. CONSTITUTION:A novel thermophilic microorganism belonging to Thermus genus and capable of producing a novel heat-resistant pullulanase, e.g. AMD-6 strain (FERM P-8881) is inoculated in a proper medium and cultured at 5.0-8.5pH and 37-74 deg.C for about 48-96hr under aerobic condition to produce and accumulate cell-adsorption-type and extracellular secretion-type novel pullulanase in the cultured liquid. Both kinds of the enzymes are separately collected or collected at the same time by adding a surfactant to the cultured liquid. |
