NOVEL DNA, NOVEL PLASMID HAVING SAID DNA, NOVEL POLYPEPTIDE AND PRODUCTION THEREOF AND NOVEL ANTITUMOR AGENT CONSISTING OF SAID POLYPEPTIDE

摘要

PURPOSE:To prepare an antitumor agent, capable of inhibiting propagation of cancerous cells, by cultivating a microorganism having a DNA capable of coding a specific amino acid sequence inserted into a plasmid propagable in host microbial cells. CONSTITUTION:A DNA capable of coding an amino acid sequence [amino acid sequence of the fourth exon part of tumor necrosis factor(TNF)] expressed by formula 1 (X represents Ser or Cys) is synthesized and integrated into a plasmid propagable in host microbial cells to form a recombinant DNA, which is then used to transform a host of a microorganism, such as animal cells, yeasts, Bacillus subtilis or Escherichia coli. The resultant transformed host microorganism is subsequently cultivated to produce a polypeptide having the amino acid sequence expressed by formula 1. An antitumor agent is prepared from the above-mentioned polypeptide having the amino acid sequence expressed by formula 1.
