PURPOSE: To measure an anti-DNA antibody value in a biological liquid accurately by preparing plasmid DNA from colitis germ without containing a single- chain DNA and cutting it with a proper limiting enzyme as a tracer material. CONSTITUTION: Ecolik12JM109 stock that has plasmid pNDPC1 is cultivated at an LB cultivation site to prepare plasmid. After the plasmid pNDPC 1 is cut to a fully short length by a limiting enzyme CfrI, its end is labeled by Klenow enzyme. The labeled DNA is melted in a water solution containing 50mM boric acid sodium, 15mMEDTA, and 0.01% NaN3 so that the final concentration reaches 0.01μCi/ml, which is used as a tracer solution.
