PRODUCTION OF TISSUE PLASMINOGEN ACTIVATION FACTOR USING NORMAL HUMAN CELL TRANSFORMED BY INTRODUCTION OF VIRUS GENE

摘要

PURPOSE:To facilitate mass-production of a tissue plasminogen activation factor in high efficiency, by introducing a vector containing SV40T antigen gene into a normal human cell and culturing the obtained transformed cell in a medium. CONSTITUTION:E.coli HB101 holding vector pSV3neo containing SV40T antigen gene is cultured and the obtained vector is linearized with a restriction enzyme. A solution of the linearized vector DNA is added to a cultured liquid suspension of normal human cell (preferably normal human diploid fibroblast cell IMR90) and the cell is transformed by introducing the linearized vector DNA into the cell according to an electropolation method. The transformed cell is cultured in a medium and the objective tissue plasminogen activation factor is separated from the cultured liquid.
