<p>Antigenic protein-hapten conjugates (immunogens) are produced in water-insoluble form from water-labile haptens, such as penicillin and other beta-lactam antibiotics, by reacting the protein and the water-labile hapten in aqueous solution under conditions, e.g. an excess of cross-linking agent, which cause substantially simultaneous precipitation of the protein-hapten conjugate before any substantial hydrolysis of the water-labile hapten has occurred. Such immunogens are used to raise antibodies, e.g. anti-penicillin antibodies, having specificity to the free, non-derivatised hapten, e.g. free non-derivatised penicillin. Such anti-penicillin antibodies are particularly useful in immunoassay of penicillin and other beta-lactam antibodies in milk.</p> |
