Proline derivs. (I)[R=acyl; A=Gly, MeGly or a D- -amino acid) or their salts were prepd. by amidation of the corresponding R-A-D- SCH2CHMeCO2H with proline or its derivs. or by esterification of R-A- OH with captopril or its salts. Thus, (I)[R=cyclohexanecarbonyl, A=D-Ala) was prepd. by condensing 3-(Ncyclohexanecarbonyl-D- alanylthio)-2-D-methylpropanic acid with proline by ClCO2Et. (I) are antihypertensives without secondary effects attributed to the mercapto group in Captopril.
