PURPOSE:To obtain a transformant in reduced dispersion for taking an extraneous DNA in, good reproducibility and high efficiency, by using bacterial spore as recipient bacterium. CONSTITUTION:Spheroplast suspension is prepared by chemically treating a bacterial spore using a bacterial spore as recipient bacterium for transformation and then treating the spore with lysozyme to remove a cell wall. An extraneous plasmid DNA is added to the resultant suspension and the mixture is shaken and kept in temperature to provide a transformant. When the plasmid DNA is introduced, the introduced efficiency can be enhanced by carrying out polyethylene glycol treatment. The transformant is readily stored and can be stored over long period and scattering by species of bacterium or experimenter is reduced by using the above-mentioned bacterium spore. |
