CELL LINE PRODUCING MONOCLONAL ANTIBODY AND MONOCLONAL ANTIBODY PRODUCED THEREFROM

PURPOSE:To obtain a monoclonal antibody useful for the detection of methamphetamine, etc., by collecting an antibody-producing cell from a mouse immunized with a protein bonded with methamphetamine or a methamphetamine derivative and cloning the cell. CONSTITUTION:A complete antigen is prepared from a substance produced by bonding hemocyanine originated from false impet (Macroschisma sinensis) and methamphetamine or a methamphetamine derivative. The antigen is mixed with Freund's complete adjuvant containing dead cell of human tubercle bacillus and an A/J mouse is immunized with the mixture. The spleen cell of the animal is extracted and fused with a cell line p3X63-Ag8.653 originated from myeloma. A cell line exhibiting high titer against methamphetamine and high proliferation property is selected from the fused cells by ELISA method. A monoclonal antibody specifically linking with methamphetamine or a methamphetamine derivative can be obtained from the cell line.
