<p>PURPOSE:To directly, rapidly and accurately assay microbial concentration in an activated sludge, i.e. activity, by measuring ATP concentration coped with the microbial concentration in activated sludge. CONSTITUTION:The activity of activated sludge to be assayed is determined from the relationship of microbial concentration of activated sludge, i.e. activity with ATP concentration and ATP concentration by obtaining the above- mentioned relationship and the ATP concentration and simultaneously measuring ATP concentration of activated sludge to be assayed and the ATP concentration is measured e.g. by collecting a sample, carrying out dilution and pretreatment by supersonic treatment, blending the pretreated sample with a tris buffer, stirring the pretreated sample in boiled water bath, extracting ATP from a miroorganism in activated sludge, adding a luminescent reagent to the centrifuged extract and measuring strength of the resultant luminescence.</p> |
